RAB14 encodes a small GTPase that regulates intracellular membrane trafficking between the Golgi complex and endosomes, particularly controlling transport of FGFR-containing vesicles during early embryonic development and mediating ADAM10 endocytic transport and N-cadherin shedding. The gene is highly intolerant to loss-of-function variation (pLI = 0.99, LOEUF = 0.21), suggesting that pathogenic variants would likely cause severe developmental disorders, though specific disease associations have not yet been established. Given its role in early development and neurite formation, mutations would be expected to follow an autosomal dominant inheritance pattern and cause developmental abnormalities through haploinsufficiency.
