GAGE10
Chr XG antigen 10
Also known as: GAGE-10
GAGE10 encodes a member of the GAGE (G antigen) family of proteins that are normally expressed in testis and placenta but can be aberrantly activated in various cancers. This gene has low evolutionary constraint with tolerance to loss-of-function variants. Currently, no definitive Mendelian disease associations have been established for GAGE10 mutations in pediatric neurological disorders.
Population Genetics & Constraint
gnomAD v4 — loss-of-function & missense intolerance
Highly tolerant — LoF variants common in population
Tolerant to missense variation
The highest-scoring mechanism for this gene is dominant-negative.
Note: In-silico variant effect predictors (SIFT, PolyPhen, REVEL, CADD) may underestimate pathogenicity of missense variants in genes with GOF or DN mechanisms. Consider functional evidence and clinical context.
